Regulatory T cells (Treg) and platelets play a crucial role in the pathogenesis of SLE by regulating immunity and promoting inflammation. Ferroptosis, an iron-regulated cell death process, is emerging as a key player in many diseases, including SLE.\n\nThe project, FERROLUP, aims to understand the role of ferroptosis in SLE and to explore the therapeutic potential of selenium compounds to modulate this process. Recent work has identified down-regulation of glutathione peroxidase 4 (GPx4) by immune complexes and interferon-alpha in neutrophils, leading to ferroptosis and worsening of SLE. In addition, data suggest the involvement of ferroptosis in lupus nephritis.\n\nThe Bordeaux team has developed selenium compounds, GPx4 mimics, capable of inhibiting ferroptosis in lupus neutrophils. These compounds have shown promising efficacy in mouse models and preliminary human studies in another inflammatory disease. The FERROLUP project aims to characterize the level of lipid peroxidation and GPx4 expression in SLE patients, and to test the impact of selenium compounds on the inhibition of ferroptosis induced by P-selectin, a molecule involved in Treg dysfunction.",[31,36],{"label":32,"type":33,"description":11,"interventionNames":34},"Systemic lupus erythematosus (SLE)","EXPERIMENTAL",[35],"Biological: blood sample",{"label":37,"type":38,"description":11,"interventionNames":39},"Rheumatoid arthritis or an inflammatory bowel","ACTIVE_COMPARATOR",[35],[41],{"type":42,"name":43,"description":44,"armGroupLabels":45,"otherNames":11},"BIOLOGICAL","blood sample","30 ml whole blood for Peripheral blood mononuclear cell (PBMC) and monocytes isolation",[37,32],[47],{"name":48,"affiliation":5,"role":49},"Christophe RICHEZ, Prof","PRINCIPAL_INVESTIGATOR",[51,56],{"name":48,"role":52,"phone":53,"phoneExt":54,"email":55},"CONTACT","(0)5 56 79 55 56","+33","christophe.richez@chu-bordeaux.fr",{"name":57,"role":52,"phone":58,"phoneExt":54,"email":59},"Thomas BARNETCHE, PhD","(0)5.57.82.04.93","thomas.barnetche@chu-bordeaux.fr",[61],{"facility":62,"status":63,"city":64,"state":11,"zip":11,"country":65,"countryCode":66,"cosmosGeoPoint":67,"geoPoint":72,"contacts":73},"CHU de Bordeaux - service de rhumatologie","RECRUITING","Bordeaux","France","FR",{"type":68,"coordinates":69},"Point",[70,71],-0.58046,44.84124,{"lat":71,"lon":70},[74,76,77,78,81,83,85,87],{"name":48,"role":52,"phone":75,"phoneExt":54,"email":55},"(0)5.56.79.55.56",{"name":57,"role":52,"phone":58,"phoneExt":54,"email":59},{"name":48,"role":49,"phone":11,"phoneExt":11,"email":11},{"name":79,"role":80,"phone":11,"phoneExt":11,"email":11},"Lionel COUZI, Prof","SUB_INVESTIGATOR",{"name":82,"role":80,"phone":11,"phoneExt":11,"email":11},"Julien SENESCHAL, Prof",{"name":84,"role":80,"phone":11,"phoneExt":11,"email":11},"Noemie GENSOUS, MD",{"name":86,"role":80,"phone":11,"phoneExt":11,"email":11},"Estibaliz LAZARO, Prof",{"name":88,"role":80,"phone":11,"phoneExt":11,"email":11},"Pauline RIVIERE, MD",{"type":90,"investigatorFullName":11,"investigatorTitle":11,"investigatorAffiliation":11,"oldNameTitle":11,"oldOrganization":11},"SPONSOR","100613004",false,"NCT07260942","Ferroptosis Role in the Pathophysiology of Systemic Lupus Erythematosus","FERROLUP","Inclusion Criteria:\n\n* age ≥ 18 years;\n* diagnosis of systemic lupus erythematosus;\n* being affiliated to health insurance, willing to participate and to sign informed consent;\n* control group : patients with a diagnosis of rheumatoid arthritis or an inflammatory bowel disease.\n\nExclusion Criteria:\n\n* pregnant or breastfeeding women;\n* patient concerned by articles L 1121-5 to L 1121-8 (persons deprived of their liberty by a judicial or administrative decision, minors, persons of legal age who are the object of a legal protection measure or unable to express their consent).","ALL","18 Years",{"count":100,"type":101},120,"ESTIMATED","INTERVENTIONAL",[104],"NA","The study aims at defining the role of ferroptosis s in the physiopathology of systemic lupus erythematosus (SLE).
