The effect of DENV or DENV-Associated Molecular Patterns (PAMPs) will therefore be studied in vitro on purified cells from SC and SS patients.\n\nThe impact of MicroParticles (MPs), known to be involved in the pathogenesis of severe dengue forms and in sickle cell disease, will also be evaluated on two cell types: endothelial cells and neutrophils, the latter exhibiting exacerbated activation and production of Neutrophil Extracellular Traps (NETs).\n\nConsidering the influence of mosquito saliva components on the pathogenesis of arboviruses (including DENV) in vertebrate hosts through the amplification of inflammatory responses, a comparative analysis of cellular and molecular inflammatory mediators released following exposure of blood cells from SS and SC patients to DENV and Aedes aegypti saliva will be conducted in vitro.\n\nLastly, the differential blood profiles of SS and SC patients will be assessed for their impact on the vector competence of Aedes aegypti mosquitoes in transmitting DENV.","patients from 6 to 25 years, with sicke cell disease in a steady state at inclusion",[29,32,35],{"label":30,"type":23,"description":31,"interventionNames":23},"Patients SS Group","Fifty SS patients",{"label":33,"type":23,"description":34,"interventionNames":23},"Patients SC","Fifty SC patients",{"label":36,"type":23,"description":37,"interventionNames":23},"Control group AA","Patients without hemoglobin disease",[39],{"name":40,"affiliation":41,"role":42},"Maryse Etienne-Julan, MD PhD","CHU de la Guadeloupe","PRINCIPAL_INVESTIGATOR",[44,49],{"name":45,"role":46,"phone":47,"phoneExt":23,"email":48},"Valérie Hamony Soter","CONTACT","+590590934677","valerie.soter@chu-guadeloupe.fr",{"name":50,"role":46,"phone":51,"phoneExt":23,"email":52},"Mélanie petapermal","+590590934667","melanie.petapermal@chu-guadeloupe.fr",[54],{"facility":55,"status":56,"city":57,"state":23,"zip":58,"country":59,"countryCode":23,"cosmosGeoPoint":60,"geoPoint":65,"contacts":66},"Chu de La Guadeloupe","RECRUITING","Pointe-à-Pitre","97159","Guadeloupe",{"type":61,"coordinates":62},"Point",[63,64],-61.53459,16.23638,{"lat":64,"lon":63},[67,71],{"name":68,"role":46,"phone":69,"phoneExt":23,"email":70},"Maryse etienne julan, MD","+590590934670","maryse.etienne-julan@chu-guadeloupe.fr",{"name":40,"role":42,"phone":23,"phoneExt":23,"email":23},{"type":73,"investigatorFullName":23,"investigatorTitle":23,"investigatorAffiliation":23,"oldNameTitle":23,"oldOrganization":23},"SPONSOR","100593000",false,"NCT07000747","Molecular and Cellular Basis of Severe Forms of Dengue in Sickle Cell Patients","DREPADENGUE","Inclusion Criteria:\n\n* patients with SS or SC SCD or controls with hemoglobin AA\n* diagnosis of SCD performed by electrophoresis or HPLC in a reference laboratory for hemoglobinopathies\n* patients older than 6 years and younger than 25 at inclusion\n* clinically in a steady state at inclusion (without complication during the last month and without transfusion in the three last months)\n* patients followed up for SCD at the sickle cell center of Guadeloupe (University hospital of Guadeloupe, Pointe-à-Pitre)\n* patients or legal representatives of minors who will provide written informed consent in accordance with the Declaration of Helsinki\n* patients affiliated to national social security\n* the control group (AA subjects) will be recruited among volunteers recruited by posters\n\nExclusion Criteria:\n\n* patients with SS or SC SCD or controls with hemoglobin AA\n* diagnosis of SCD performed by electrophoresis or HPLC in a reference laboratory for hemoglobinopathies\n* patients older than 6 years and younger than 25 at inclusion\n* clinically in a steady state at inclusion (without complication during the last month and without transfusion in the three last months)\n* patients followed up for SCD at the sickle cell center of Guadeloupe (University hospital of Guadeloupe, Pointe-à-Pitre)\n* patients or legal representatives of minors who will provide written informed consent in accordance with the Declaration of Helsinki\n* patients affiliated to national social security\n* the control group (AA subjects) will be recruited among volunteers recruited by posters",true,"ALL","6 Years","25 Years",{"count":85,"type":86},130,"ESTIMATED","OBSERVATIONAL","Dengue virus (DENV) belongs to the genus of Flavivirus transmitted by the mosquito Aedes aegypti and is responsible for an infectious disease associated with different forms and severities such as dengue hemorrhagic fever or shock syndrome. Several recent reports have shown that sickle cell patients exhibited an increased risk of developing severe forms of dengue episodes compared to non-sickle cell subjects. Furthermore, among major sickle cell syndromes, these studies suggest that SC patients are at the highest risk of death during these infectious episodes although this sickle cell syndrome is generally associated with a more moderate expression of sickle cell disease. However, the mechanisms involved remain unknown to date.\n\nThe aim of the present study is to identify the molecular and cellular basis of this increased severity of dengue in SC patients.
