This knowledge gap represents a relevant unmet need in oncofertility counseling and survivorship care.\n\nThe study will include patients diagnosed with stage I-III invasive breast cancer at age 40 years or younger between January 2000 and December 2025. The primary objectives are to assess the prognostic impact of pregnancy after breast cancer diagnosis and to evaluate the cumulative incidence of pregnancy in this population. Secondary objectives include the assessment of pregnancy, fetal, and obstetrical outcomes, the safety of assisted reproductive technology procedures, patterns of care including risk-reducing surgeries, survival outcomes according to clinicopathologic and genetic subgroups, and the occurrence of second primary malignancies.\n\nData will be collected retrospectively from participating centers within the Beyond BRCA BCY Collaboration.","Young women diagnosed at age 40 years or younger with stage I-III invasive breast cancer between January 2000 and December 2025 and harboring a germline pathogenic variant in a breast cancer susceptibility gene other than BRCA1\u002F2, including TP53, PALB2, PTEN, CDH1, STK11, CHEK2, ATM, BARD1, RAD51C, and RAD51D.",[21,24],{"label":22,"type":11,"description":23,"interventionNames":11},"Pregnant cohort","Women with one or more pregnancies any time after breast cancer diagnosis",{"label":25,"type":11,"description":26,"interventionNames":11},"Non-pregnant cohort","Women with no subsequent pregnancies after breast cancer diagnosis",[28,32],{"name":29,"affiliation":30,"role":31},"Matteo Lambertini, MD, PhD","IRCCS Azienda Ospedaliera Metropolitana","PRINCIPAL_INVESTIGATOR",{"name":33,"affiliation":5,"role":31},"Evandro de Azambuja, MD, PhD",[35,40],{"name":36,"role":37,"phone":38,"phoneExt":11,"email":39},"Eva Blondeaux, MD","CONTACT","+39 010 555 8502","BCYcollaboration@aomliguria.it",{"name":41,"role":37,"phone":11,"phoneExt":11,"email":39},"Luca Arecco, MD",{"type":43,"investigatorFullName":11,"investigatorTitle":11,"investigatorAffiliation":11,"oldNameTitle":11,"oldOrganization":11},"SPONSOR","100638324",false,"NCT07591532","Pregnancy Outcomes After Breast Cancer in Young Women With Germline Pathogenic Variants in Genes Other Than BRCA","Retrospective Observational Study on the Prognostic Impact of Pregnancy in Young Women With Breast Cancer Harboring a Germline Pathogenic Variant in Breast Cancer-related Genes Other Than BRCA1\u002F2: the \"Beyond BRCA BCY Collaboration\"","Beyond BCY","Inclusion Criteria:\n\n* Diagnosis of stage I-III invasive breast cancer between January 2000 and December 2025\n* Age at breast cancer diagnosis ≤40 years\n* Germline pathogenic variant in at least one of the following breast cancer susceptibility genes other than BRCA: TP53, PALB2, PTEN, CDH1, STK11, CHEK2, ATM, BARD1, RAD51C, or RAD51D\n\nExclusion Criteria:\n\n* Known germline pathogenic variant in breast cancer susceptibility genes without a diagnosis of invasive breast cancer\n* Diagnosis of ovarian cancer or other malignancies without a prior history of invasive breast cancer\n* Diagnosis of invasive breast cancer with germline variants of uncertain significance in breast cancer susceptibility genes\n* De novo stage IV breast cancer","FEMALE","18 Years","40 Years",{"count":55,"type":56},2200,"ESTIMATED","OBSERVATIONAL","The present study aims to refine the understanding of the prognostic impact of pregnancy after breast cancer in young women harboring germline pathogenic variants in breast cancer susceptibility genes other than BRCA",[60,61],"Breast Cancer","Breast Cancer and Pregnancy",[63,64,65,66,67,68,69,70,71,72,73,74,75,76,77,78,79],"breast cancer","Pregnancy","Young Women","Hereditary Breast Cancer","Germline Pathogenic Variants","Breast Cancer Susceptibility Genes","Oncofertility","TP53","PALB2","PTEN","CDH1","STK11","CHEK2","ATM","BARD1","RAD51C","RAD51D","NOT_YET_RECRUITING","2026-05-11",{"date":83,"type":84},"2026-05-15","ACTUAL",{"date":86,"type":56},"2026-06",{"date":88,"type":56},"2030-12",{"name":5,"class":6}]
